摘要
目的探讨采用超临界流体CO2萃取技术提取龙胆中龙胆总苷的工艺。方法应用超临界流体CO2对龙胆总苷进行提取,在提取过程中加入体积分数为95%的乙醇作为夹带剂,用紫外-可见分光光度法(UV-2450)测定其总苷含量。结果在萃取压力25 MPa,萃取温度50℃,分离压力6 MPa,分离温度55℃的条件下,所得的提取物中总苷量最高。结论通过加夹带剂的方法,应用超临界流体CO2萃取技术可提取出龙胆中极性较大的苷类成分。
Objective To establish a method for extracting the glycosides of Radix Gentianae. Methods Supercritical CO2-ethanol was used to extract the glycosides. The content of total glycosides was measured by UV. Temperatures, pressures and concentrations of ethanol (modifier) and the ratio (w/w) of solvent to material were tested in this study. Results Twenty-five MPa of extraction pressure,50 ℃ of extraction temperature,6 MPa of separation pressure and 55℃ of separation temperature, were the most favorable extraction conditions. Conclusion The glycosides could be effectively extracted from Radix Gentianae by supercritical fluid extraction with CO2-ethanol.
