TPO和FL有效扩增脐血CD34+细胞为巨核前体细胞 被引量:1

摘要 目的探讨人脐血CD34+细胞在TPO和FL细胞因子组合作用下体外扩增为巨核前体细胞的效果,为开展血小板减少症的细胞治疗建立实验条件。方法本研究采用人脐血经免疫磁珠法纯化分离的CD34+细胞,研究TPO+FL细胞因子组合对体外巨核前体细胞的扩增效果。结果显示在TPO与FL组合的作用下,脐血CD34+细胞(n=6)显著地扩增为巨核前体细胞(CD61+CD41+细胞),作用14天为较理想时间。无论是全细胞数和CD34+细胞,或是CD61+CD41+细胞和CFUMK,经过体外扩增,均显著增加。结论研究表明脐血CD34+细胞在TPO和FL细胞因子组合作用下可有效地扩增为巨核前体细胞,第14天是培养较理想时间。 Objective To investigate the effects of TPO in combination with FL on ex vivo expansion of magakaryocytic progenitors. Methods Mononuclear cells (MNC) were separated from human umbilical cord blood samples (n = 6) , and CD34 + stem and progenitor cells were purified by using immunomagnetic beads, Numbers of nucleated cells, CD34 + cells, CD34 + CD38-cells, CD41 + CD61 + cells and CFU-MK were evaluated at 7th, 14th and 21th days after culture, effects of TPO in combination with FL were investigated on the differentiation and proliferation of megakaryocyte progenitor cells. Results Enriched CD34 + stem and progenitor cells were significantly expanded in the presence of TPO combined with FL. There were overall increases of nucleated cells, CD34 + cells, CD34 + CD38-cells, and CD61 + CD41 + cells as well as CFU-MK. The culture for 14 days appeared to be the most effective duration of expanding all cell types studied. Conclusion It is showed that the mixture of TPO and FL could significantly promote the ex vivo expansion of megakaryocytic progenitors from CB CD34 + hematopoietic cells, and the culture for 14 days appeared to be the optimal duration of expansion.
