摘要
探讨并构建了三酶复合法提取大豆多肽的新工艺。即将大豆分离蛋白经三酶复合酶解、活性炭脱色、超滤、真空浓缩和离心喷雾干燥等提取工艺,最终获得大豆多肽。其制备的关键工艺条件为①最佳复合酶解条件:中性蛋白酶、木瓜蛋白酶、菠萝蛋白酶比例为1∶2∶4,pH 7.0,底物浓度4.0%,酶解时间9 h;②最佳脱色处理条件:粉末活性炭用量2.0%,温度50℃,pH 3.0,吸附时间3 h;③超滤分离条件:NF-1纳滤膜,压力1.0 MPa,膜通量26.0 L/(m2.h)。
The new extraction technique of soybean pepfides by triple-enzyme composite hydrolysis was established in this study. Soybean peptides were obtained from isolated soybean protein by triple-enzyme composite hydrolysis and purification. The result showed that the best conditions of composite enzymatic hydrolysis were: the composite enzymes were made up of neutral proteinase, papain and bromelain with the ratio of 1:2:4,pH 7.0,4.0% of substrate concentration, and hydrolysis time for 9 h; the best conditions of decoloration were: 2.0% of activated carbon, 50℃ of temperature, pH 3.0, adsorption time for 3 h; the ultra-filtration conditions were: NF-1 nanometer membrane, 1.0 MPa of pressure and 26.0 L/(m^2·h) of the permeate flux.
