摘要
目的探讨逆灸关元穴对自然更年期大鼠子宫结构、子宫内激素和受体的影响及保护机制。方法将128只SD雌性大鼠随机分8组,采用免疫组化、放射免疫、HE染色等方法,观察在10月龄逆灸对随后12、14、16月龄大鼠子宫的组织形态及激素和受体的影响。结果随增龄,模型对照各组大鼠子宫的结构出现明显退化,同时子宫内激素雌二醇(E2)、孕激素(P)的含量同步下降。而从10月龄介入逆灸关元方法后,各组子宫与同期对照组比较,情况明显好转;逆灸还可提高子宫内E2及受体表达,E2含量的提高主要在14、16月龄(均P<0.05);雌激素受体(ER-α)表达的加强则主要在12、14月龄组(P<0.01,P<0.05);逆灸还可调节子宫内P的含量,其效应主要体现在16月龄(P<0.05)。结论逆灸关元穴对更年期大鼠子宫退行性改变具有延缓作用,这种保护作用可能与逆灸调节子宫局部E2、P及ER-α表达有关。
Abstract:Objective To investigate the influence and protective mechanism of routine moxibustion on the acupoint of Guanyuan (CV4) on uterus structure, hormones and receptor in natural climacterium rats. Methods To divide randomly 128 SD female rats into 8 groups. The changes of histomorphology, hormones and receptor in uterus were observed by using immunohistochemistry, radioimmunoassay and HE staining in 12^th, 14^th and 16^th month after routine moxibustion was given to rats aged ten months. Results There was obvious catagenesis in rat uterus sructure in all model control groups and the contents of estradiol (E2 ), progestagen (P) in uterus were decreased at the same time. After the routine moxibustion on CV4 in rats aged 10 months the catagenesis of uterus in different groups relieved compared with that of the control group. Routine moxibustion improved the expressions of E2 and receptor in uterus. The increase of E2 content was mainly observed in 14^th and 16^th month groups (all P 〈 0. 05 ). The increase of ER-α expression was mainly observed in 12^th and 14^th month group (P 〈0. 01, P 〈0. 05). Routine moxibustion had regulated P content in uterus, which was mainly observed in 16^h month group (P 〈 0.05 ). Conclusion Routine moxibustion on CV4 can prevent climacterium rat from retrograde changes of uterus. The effect may be related to that routine moxibustion can regulate the contents of E2 , P and ER-α expression in uterus.
