摘要
以百合叶片为材料,建立了百合叶片不定芽发生、植株再生和微繁的技术体系。研究了不同消毒时间、不同激素及其组合对叶片不定芽发生、增殖和生根的效应。结果显示,用0.1%的氯化汞对叶片消毒5 min效果较好,污染率为36%,萌动率为75%,诱导率为68.8%。MS+6-BA 2.0 mg/L+NAA 0.5 mg/L是叶片不定芽发生的适宜培养基;MS+6-BA 1.0 mg/L+NAA 0.1 mg/L是不定芽增殖的适宜培养基;MS+NAA 0.1 mg/L是生根的适宜培养基。
Adventitious shoot formation, plant regeneration and micro-propagation were established in lily leaf in vitro. Effect of diffen;nt sterilized time, kinds of hormone and its combination on adventitious shoot formation, plant regeneration and root regeneration were studied. The results showed that explant sterilized in 0.10 % aqueous mercuric chloride for 5 rain 'had better effect,and 36 % of coutaminatiou rate,75 % of germinating rate, 68.8 % of inducement rate, MS + 6- BA 2.0 mg/L + NAA 0.5 mg/L was suitable for adventitious shout formation from the leaf, the sheots grew well in MS + 6- BA 1.0 mg/L + NAA 0.1 mg/L medium, MS + NAA 0.1 mg/L was the best medium for root regeneration.
