摘要
目的探讨沙眼衣原体(CT)K血清型对细胞凋亡的影响。方法用流式细胞仪(FCM)检测细胞凋亡百分率,DNA琼脂糖凝胶电泳检测细胞凋亡“梯状带”,观察沙眼衣原体K血清型感染36h,48h,60h对BGM细胞和Mc-Coy细胞凋亡的影响。结果沙眼衣原体感染的BGM细胞在36h,48h,60h的细胞凋亡百分率分别为(3.41±0.09)%,(8.96±0.78)%和(16.60±0.41)%,沙眼衣原体感染的McCoy细胞在36h,48h,60h的细胞凋亡百分率分别为(2.63±0.11)%,(8.78±0.58)%和(12.93±0.45)%,均显著高于相应的对照组(P<0.01),且凋亡百分率随时间延长而增加(P<0.01)。结论沙眼衣原体K血清型能诱导细胞凋亡。
Objective To investigate the effect of Chlamydia trachomatis serotype K infection on apoptosis. Methods Quantitative measurement of apoptosis and detection of "DNA ladders" were performed by FCM and agarose electrophoresis respectively in BGM cells and McCoy cells infected with Chlamydia trachomatis at different time (36 h ,48 h ,60 h). Results DNA ladder were observed in BGM cells and McCoy cells from 48 h to 60 h after infected with Chlamydia trachomatis K serotype, The percentage of apoptotic cells in BGM cells at different time after infected [ (3.41 ±0.09 ) %, ( 8.96 ±0.78 ) %, ( 16.60 ±0.41 ) % respectively] were much higher than that of the corresponding control groups [ ( 1.26 ± 0.02 ) % , ( 1.56 ± 0.06 ) % , ( 2.04 ±0.04) % respectively P 〈0.01 ]. Chlamydia trachomatis K serotype induced obvious apoptosis of McCoy cells with apoptosis ratio of( 2.63 ±0.11 ) % , ( 8.78 ± 0.58 ) % , ( 12.93 ±0.45 ) % ascomparedwith thecontrolgroups [ ( 1.17 ±0.03 ) % , ( 1.30 ±0.04) %, ( 1.97 ±0.46 ) % respectively. P 〈 0.01 ]. The perecentage of apoptotic cells increased with the time ( P 〈 0.01 ). Condusion Infection of chlamydia trachomatis serotype K can induce BGM cells and McCoy cells apoptosis in a time dependent manner.
