摘要
目的:探讨在人结肠癌耐药株LoVo/Adr细胞中可溶性耐药相关钙结合蛋白(solubleresistancerelatedcalciumbindingprotein,sorcin)与Ca2+浓度的关系。方法:以Fluo3/AM标记细胞内Ca2+,用共聚焦显微镜观察其浓度变化;采用RTPCR和免疫组化的方法检测细胞中sorcin基因的mRNA水平和蛋白水平。结果:与亲本LoVo细胞相比,耐药LoVo/Adr细胞中Ca2+浓度显著增高(P<0.01);sorcin基因在LoVo细胞中几乎不表达,而在耐药LoVo/Adr细胞中,无论mRNA水平还是蛋白水平均显著高表达(P<0.01)。结论:耐药LoVo/Adr细胞中sorcin高表达没有引起细胞内Ca2+浓度的降低。
Objective:To investigate the relationship between soluble drug resistance-related calcium-binding protein (sorcin) gene expression and intracellular calcium concentration in multidrug resistant human colon carcinoma cell line LoVo/Adr. Methods:Intracellular calcium was labeled by Fluo-3/AM. The changes of intracellular calcium concentration were observed by confocal microscope. Sorcin mRNA and protein expression were detected by RT-PCR and immunohistochemical method, respectively. Results:The intracellular calcium concentration in LoVo/Adr cells was significantly higher than that in LoVo cells(P〈 0.01). No sorcin mRNA expression was detected in the parental LoVo cells. The expressions of sorcin were higher at both mRNA and protein level in multidrug resistant LoVo/Adr cells(P〈0.01). Conclusion: Higher expression of sorcin does not induce decrease in intracellular calcium concentration in multidrug resistant LoVo/Adr cells.
