摘要
目的检测Bclxl/LipofectamineTM2000复合物中两者比值对转染人角膜基质细胞转染效率的影响,探讨其用于人角膜基质细胞基因转染的最佳比值。方法选择Bclxl/LipofectamineTM2000比值1∶2、1∶4、1∶8、1∶10四个实验组制备载体复合物转染人角膜基质细胞,转染时间24h。在转染后3d,采用半定量RTPCR检测目的基因Bclx1mRNA表达的变化;采用免疫组织化学染色法及流式细胞计数法检测目的基因Bclx1表达阳性细胞率。数据采用SPSS软件分析。结果Bclxl/LipofectamineTM2000载体复合物中两者比值能显著影响其对人角膜基质细胞的转染效率,存在最佳的比值。当Bclxl/LipofectamineTM2000比值为1∶8时,培养的人角膜基质细胞获得了最佳的转染效率。转染后3d,定量PCR显示1∶8组mRNA表达水平高于其他组(P<0.05),Bclx1免疫组化染色及流式细胞计数法均显示1∶8组转染效率最高,阳性细胞率为(45.6±2.0)%(免疫组织化学染色法),(48.3±3.0)%(流式细胞计数法)(P<0.05)。结论LipofectamineTM2000能有效介导外源基因Bclxl转染人角膜基质细胞。Bclxl/LipofectamineTM2000载体复合物比值为1∶8时转染效率最高。
Objective. To evaluate the best ratio of Bcl-xl/Lipofectamine^TM2000 for human keratocyte transfection. Methods Different concentrations of Bcl-xl and Lipofectamine^TM2000 was mixed to make Bcl-xl/Lipofectamine^TM2000 complex, with the final Bcl-xl/LipofectamineTM2000 ratios of 1:2, 1:4,1:8 and 1 : 10 respectively. All the Bcl-xl/Lipofectamine^TM2000 complex were incubated with cultured human keratocyte for 24 hours before transfection. The transfectton efficiency was tested with RT-PCR, immunohtstochemtstry, flow cytometry methods on the 3rd day of transfection. The data was analyzed with SPSS software. Results The transfection efficiency of Bcl-xl gene was various dependent on the Bcl-xl/Lipofectamine^TM2000 ratio. Bcl-xl/Lipofectamine^TM2000 ratio was 1 : 8, the highest transduction efficiency appeared as high as (45.6 ± 2.0)% (tmmunohtstochemtstry), (48.3 ± 3.0)% ( flow cytometry) ,significantly higher than other groups(P〈0.05). Conclusion Under appropriate conditions, cationic liposome can facilitate the transfection of Bcl-xl gene into keratocyte. The best ratio of Bcl-xl/Lipofectamine^TM2000 was 1 : 8.
