异硫氰酸丙烯酯对大鼠肝细胞凋亡的影响 被引量:1

摘要 为研究异硫氰酸丙烯酯对细胞活性的影响,试验以20mg·L-1的内毒素(LPS)和1.25×10-5mol·L-1的异硫氰酸丙稀酯(AITC)分别或共同处理大鼠肝细胞3h后,用AnnexinV/PI双染色,流式细胞术分析细胞凋亡;反转录酶聚合酶链反应(RT-RCR)分析FasL和Caspase-8的表达情况。结果表明,AITC能有效抑制LPS(20mg·L-1)诱导的肝细胞凋亡,且在此过程中,FasL、Caspase-8的表达量均有减少。结果提示AITC可以通过Fas途径抑制LPS诱导的肝细胞凋亡,其作用有可能导致肿瘤的发生。 In order to study the effect of allyl isothiocyanate (AITC) towards rat hepatocytes' activity, LPS(20 mg·L^-1) and AITC 1.25×10^-5 mol·L^-1 were used partly or together to act on cultured hepatocytes. After 3 hours, apoptosis was detected by Annexin V/PI staining and flow cytometry, accompanied by the change of FasL, Caspase-8 expression analyzed by PT-PCR. The result showed that AITC had the right to reduce the rate of rat hepatocytes apoptosis caused by LPS (20 mg·L^- 1) and the expression of Caspase-8 ,FasL. It suggested allyl isothiocyanate have the potential to induce the turnout and cancer when spontaneous apoptosis of the two kinds of cell be restrain.
