白松片对慢性应激大鼠海马5-羟色胺和多巴胺含量的影响 被引量:1

Effect of Bai-song Tablet on 5-hydroxytryptamine and Dopamine in Hippocampus of Chronic Unpredictable Stress Rat Model

摘要 目的观察中药白松片对慢性应激大鼠海马5-羟色胺(5-HT)和多巴胺(DA)含量的影响。方法健康成年雄性SD大鼠42只,随机分为正常对照组、模型对照组、盐酸氟西汀对照组(1.8mg·Kg-1)及白松片3个剂量(4.32、13.0、21.6g·Kg-1,生药量)组。每只大鼠每日灌胃给药1次,连续14d。给药d6始,通过强迫游泳建立应激大鼠模型。用高效液相色谱-电化学法测定大鼠海马5-HT和DA。结果慢性应激模型组大鼠海马5-HT和DA含量低于正常对照组(P<0.01);用白松片预防给药可使模型大鼠海马5-HT和DA含量恢复至正常水平(P<0.05或P<0.01)。结论提示白松片能提高海马5-HT和DA的含量是其抗抑郁作用机制之一。 Objective To investigate the effect of Bai- song tablet on 5 - hydroxytryptamine (5 - HT) and dopamine (DA) in hippocampus of experimental chronic unpredictable stress rats. Methods Forty- two adult male Sprague- Dawley rats were randomly divided into six groups: the normal control group, model control group, fluoxetine hydrochloride (1.8 mg.Kg^-1 ) control group and three different doses (4.32, 13.0, 21.6 g·Kg^-1, crude drug) of Bai-song tablet groups. The drug and placebo were administrated once every clay through gastric tube for 14 successive clays. The stress rat models were established by forced swimming during the 6th experimental clay. The concentrations of 5 - HT and DA in rat hippocampus were assayed by high performance liquid chromatography coupled with electrochemical detection by the end of experiment. Results The levels of 5 - HT and DA in hippocampus of chronic unpredictable stress rat model were significantly lower than that of the normal control rats ( P〈 0.01 ). The Bai - song tablet can normalize the levels of 5 - HT and DA in hippocampus of model rats (P〈 0.05 or P〈 0.01). Conclusion The results suggest that Bai - song tablets exhibit the effect of anti - depression through upregulating the levels of 5 - HT and DA.
