摘要 目的:研究cyclinD1和p16在大肠癌中的表达及临床意义。方法:应用流式细胞术(FCM)方法测定大肠癌和正常对照组结肠标本的cyclinD1和p16基因蛋白,蛋白表达量以荧光指数(fluorescentindex,FI)表示。结果:大肠癌组织cyclinD1、p16基因蛋白的FI值与正常大肠黏膜组织比较差异有显著性(P<0.01,P<0.02)。CyclinD1、p16的异常表达与肿瘤组织学类型、Duke′s分期、淋巴结转移密切相关(P<0.01)。结论:大肠癌的发生与cyclinD1高表达和P16低表达均有关,cyclinD1和p16基因可作为大肠癌早期诊断、临床生物学行为和预后评估的重要指标。 Objective:To investigate the expressions and clinical significance of cyclinD1 and P16 in colorectal carcinoma. Methods :CyclinD1 ,P16 of colorectal carcinoma and normal large intestine tissue specimen were measured using flow cytometry technique(FCM). The expression of protein was expressed by fluorescent index (FI). Results :The differences were significant between the colorectal carcinoma and normal tissue groups in FI of genes. The expression of cyclinD1 and p16 were significantly related with histoligical differentiation,Duke's stage and lymph node metastasis(P〈 0. 01). Conclusions:The occurrence of colorectal carcinoma is related to high expression of cyclinD1 and low expression of p16. CyclinD1 and p16 may be considered as prediction of diagnosis,clinical biological behavior and prognosis of colorectal carcinoma.
