摘要
目的黄绿青霉素的合成。方法黄暗青霉菌接种于已高压灭菌的大米中混匀,培养数日后,将霉变大米的乙醇提取液,旋转蒸干,溶于苯,浓缩后得到的粗毒素在硅胶柱上层析,依次用苯、乙醚、乙酸乙酯、丙酮洗脱,收集乙酸乙酯部分浓缩,4℃放置2 d后结晶。结果所得产物经高效液相色谱检测成分单一,纯度高。结论用大米做培养基产量高,杂质少;新的柱层析法易分离,利结晶。
Objective The production of citreoviridin. Methods Innoeulate sterilized flee with Penieilliun Citreo - Viride and mix them together. Cultivate for several days then extract the moldy riee with ethanol.The extract is dissolved with benzene then concentrate it at 60℃. The crude toxin is added to a Silica Gel colume. The colume is eluted sequentially with benzene -ethyl- ethyl acetate- acetone. The ethyl acetate fraction is reduced in volume, and the toxin crystallized after 2 days at 4℃. Results The production is proved single and pure by HPLC. Conclusions Rice as culture can get higher yield and fewer impurity;the new method of coluran chromatography is easier to separate and benefit crystallization.
