摘要
以日本多花紫藤种子无菌播种小苗茎及落地苗带腋芽茎段为外植体,研究了不同时期在不同培养条件下分生组织的诱导发育情况。结果表明:初代培养的适宜培养基为MS+BA2mg/L+NAA0.1mg/L+3.5%蔗糖;继代增殖以光强1500 ̄2000Lx,温度(25±1)℃,pH5.8 ̄6.0,培养基以MS+BA2mg/L+IBA0.5mg/L+3%蔗糖增殖率最高,25 ̄30d即可转接继代,每代增殖倍数为2.8 ̄3.6倍;生根培养以1/2MS+ABT0.5mg/L+IBA0.5mg/L+2.5%蔗糖在光强1500 ̄2000Lx,温度(25±1)℃条件下,生根率达95%以上。同时,对组培苗假植基质也进行了研究筛选,以河砂+草炭+谷壳灰=2:1:1处理效果较好,移栽成活率达86.5%。
Seedling stems with lateral buds were used as explants to study the shoot induction and propagation in Japanese W. foribumda under different tissue culture condition. An efficient plant regeneration system from explants of Japanese wisteria has been established in this study. The optimal medium for the induction of multiple shoots at the first stage of culture is MS medium supplemented with BA2mg/L, NAA0.1mg/L, and 3.5% sucrose. High frequency of further multiplication were obtained in the medium of MS containing BA2mg/L, IBA 0.Smg/L, and 3% sucrose (pH 5.8~6.0), under cool white fluorescent light of 1500~2000Lx at (25±1)℃. The proliferation ratio was 2.8~3.6 at intervals of 25-30 days. The rooting rate in the medium of 1/2 MS supplemented with ABT0.Smg/L, IBAO.Smg/L, and 2.5% sucrose could reach over 95%. The regenerated plantlets were transplanted successfully in mixture soil with sand: peat: husk ashes (2: 1:1) with survival rate of 86.5%.
