富硒灵芝中高抗氧化活力、高硒含量的水溶性硒蛋白的纯化 被引量:10

Isolation of Water Soluble Selenoprotein with High Se Content and High Antioxidant Activity from Se-enriched Ganoderma Lucidum

摘要 在富硒灵芝蛋白的各种溶剂提取物中,水溶性蛋白是其中结合硒含量最高的。本文以高硒(Se)含量、高自由基清除能力为考查指标,通过优化的柱层析等方法对富硒灵芝中的水溶性硒蛋白进行了提取、分离和纯化。通过电感偶合等离子体发射光谱法(ICP-AES)测定蛋白质结合的硒元素(se)含量,采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳法(SDS-PAGE)进行纯度鉴定。通过电子自旋共振捕捉(EPR)技术测定样品对羟自由基和超氧自由基的清除能力。研究结果表明,纯化后的硒蛋白显示出很强的羟自由基和超氧自由基的清除能力,并且这种抗氧化能力随着硒蛋白中硒含量的增加而增强,从而反映了硒元素在富硒灵芝蛋白抗氧化活力提高方面的重要作用。 Water-soluble protein components extracted from Se-enriched Ganoderma lucidum (Se-GL) was found to contain the highest amount of Se compared withany other reagent - extracted proteins from Se-GL. A kind of seleno-protein, which has relatively higher selenium content and antioxidant activity, was obtained from water-~luble protein components by using optimized chromatography programs. Selenium content was determined by ICP-AES and antioxidant activity was determined by electron paramagnetic resonance (EPR) in conjunction with spin trapping techniques. The results showed that the puritied selno-protein had superoxide and hydroxyl radical scavenging activity and its activity increased with the increase of Se content. This indicated that Se plays an important role in increasing the antioxidant activities of the protein extracts.
