摘要
对从四川卧龙中国保护大熊猫研究中心提供的野外放归大熊猫“祥祥”的粪样中分离到的产纤维素酶菌株JF-1116进行摇瓶产酶培养。试验结果表明,以0.5%羧甲基纤维素钠(CMC-Na)为碳源,1%蛋白胨为氮源,3%麸皮为添加物,在28℃,20%装液量,摇床振荡培养24h后,其发酵液滤纸酶活(FPA)可达111.5U/mL。通过对其粗酶液的性质进行测定,该菌所产纤维素酶粗酶液中的FPA、CMCase和β-葡萄糖苷酶的最适产酶温度分别为50、50、55℃。其中CMCase在pH值4.2~5.4酶活较高,其最适反应pH值为4.8,经50℃处理1h后,可保持原活力的93.5%。
A bacillus strain JF-1116,afforded by China Giant Panda Protection and Research Center,Wolong Sichuan province, was isolated from the dejecta of the Giant Panda "Xiangxiang"returned to wild.By primary research,the results in shaking flask culture showed that the fermentation broth was loaded in 250 mL Erlenmeyer flasks containing 50 mL of synthetic medium consisting of CMC-Na 0.5 % ,peptone 1% ,bran 3 % and incubated at 28℃ for 48 h.Then the maximum yield of FPase was 111.5 U/mL.By further study on the character of the crude enzyme,the optimum temperature of FPase,CMCase and β-glu-cosidase activities in the crude enzyme were 50,50 and 55 ℃ respectively.The CMCase was very active over the pH value of 4.2 to 5.4 and the optimum pH value was 4.8.Compared with the primary enzyme activity,the disposed enzyme activity could keep 93.5 % at 50 ℃ for 60 min.
