摘要
目的:探寻建立真皮类似物的培养方法。方法:原代培养人皮肤成纤维细胞,制备鼠尾胶原,将鼠尾胶原溶液、浓缩培养基、NaOH溶液和以血清重悬的成纤维细胞溶液在适当的比例下混合后形成凝胶构建真皮类似物。结果:形成的真皮类似物中成纤维细胞生长状态良好,并且组织块具有一定弹性和抗拉伸能力。结论:①利用鼠尾胶原可以构建真皮类似物,该模型是进行皮肤器官型培养和制作复合皮的关键与基础;②成纤维细胞胶原凝胶复合物有着良好的生物学特性,它是研究成纤维细胞与细胞外基质之间以及细胞之间相互作用的良好模型。
Objective To establish a method of constructing a living dermis equivalent in vitro. Methods The human fibroblasts were primarily cultured and the collagen of the rat tail was also prepared. Rat tail collagen, concentrated culture medium,sodium hydroxide,the fibroblast suspension in calf serum were mixed pro rata to shape collagen gel and form dermis equivalent. Results The collagen gel made fibroblasts have a suitable condition and the fibroblast collagen gel can be stretched slightly without being torn or deformed. Conclusion①Using the rat tail collagen can form dermis equivalent, which was the key to provide the artificial skin, ②Based on the favorable biological traits of the fibroblasts in collagen gel, fibroblast collagen gel is a better model in the research of the fibroblasts interact with extracellular matrix and cells.
