摘要
为了提高基因枪法在小麦转基因中的转化效率,对培养基、筛选剂浓度和轰击距离等因素进行了研究。结果表明,采用M S、2M S和B5三种培养基对东农7742、龙6239、龙辐麦3号、龙辐麦8号、龙辐麦10号、龙麦26的幼胚进行组织培养时,最佳培养基是M S培养基。采用PPT对龙辐麦8号、龙辐麦3号的愈伤组织进行筛选时,龙辐麦8号的适宜筛选浓度为5 m g/L,龙辐麦3号为10 m g/L;用G 418对龙麦26、龙辐麦3号的愈伤组织进行筛选时,适宜的筛选浓度为25 m g/L。此外,用东农7742、龙辐麦3号、龙6239和龙辐麦10号的幼胚愈伤组织为靶材料进行基因枪轰击时,轰击距离3、6、12 cm与9 cm之间分化率的差异均达到极显著水平,其适宜的轰击距离为9 cm。
To improve the efficiency of wheat transformation, some important factors in the transformation system such as suitable medium, concentration of the selective agent and bombardment distance were studied. The results were as follows: the best medium was MS in the immature embryo culture for materials Dongnong 7742, Long 6239,Longfumai 3, Longfumai 8, Longfumai 10 and Longmai 26. The optimum concentration of selective agent PPT for callus of Longfumai 8 and Longfumai 3 was 5 mg/L and 10 mg/L, respectively. The optimum concentration of selective agent G418 for callus of Longmai 26 and Longfumai 3 was 25 mg/L. The optimum distance between DNA-gold pellet and the target cells was 9 cm.
