摘要
目的:动脉粥样硬化(Atherosclerosis,AS)斑块不稳定是导致心脑血管事件的关键因素,目前尚缺乏有效救治措施。本文主要探讨三七总皂苷(TotalSaponinofPanaxnotoginseng,PNS)对斑块稳定性的影响,并检测斑块中血管内皮生长因子(VEGF)、基质金属蛋白酶2(MMP2)的表达,以期为寻找有效改善AS斑块稳定性的药物提供实验依据。方法:高胆固醇饮食建立兔AS病变模型,取主动脉行组织形态学观察,用“ImageProPlus4.5”软件测定斑块面积、内膜总面积、内膜厚度、中膜厚度、纤维帽厚度和斑块内脂质含量及采用免疫组织化学方法对斑块组织中MMP2,VEGF进行分析。结果:PNS治疗组斑块面积、内膜厚度、脂质含量低于模型组,纤维帽厚于模型组;PNS治疗组斑块组织中MMP2,VEGF的表达显著低于模型组。结论:PNS对AS斑块有稳定作用,其机制可能与抑制VEGF,MMP2表达有关。
AIM: The instability of atheroselerotic plaque was the critical factor inducing heart events, and effective remedies for this are deficient at present. Rabbit atherosclerotic models were set up to study the effects of total saponin of Panax notoginseng (PNS) on stability of atherosclerotic plaques, expression of vascular endothelial growth factor and matrix met.alloproteinase-2 in plaques were detected by i mmunohistoehemistry, which provided evidence for searching effective drug for amending stability of atheroselerotic plaque. METHODS : Rabbit atheroselerotic models were set up by hypercholestermia diet. Total areas of endomembrane and plaque, thickness of endomembrane and tunica media, contents of lipid were all measured by Image Pro Plus 4.5. Contents of vascular endothelial growth factor and matrix metalloproteinase- 2 in plaque were detected by immunohistoehemical method. RESULTS: In PNS group, area of plaque was smaller, endomembrane and fibrous cap were thinner compared with the control group. Contents of vascular endothelial growth factor and matrix metalloproteinase-2 and lipid tound in PNS group decreased, but thickness of fibrous cap increased. CONCLUSION: PNS could enhance stability of atherosclerotic plaque by downregulating expression of vascular endothelial growth factor and matrix metalloproteinase-2 in plaques.
