摘要
采用大鼠脑缺血再灌注模型,观察系列剂量的白藜芦醇甙(Polydatin,Pol)对大鼠大脑皮层及海马部位的脑含水量、丙二醛、GSH含量及超氧化物歧化酶(Super-oxide dismutase,SOD)、过氧化氢酶(Catalase,CAT)、谷胱苷肽过氧化物酶(Glutathione Peroxidase,GSH-Px)活性的影响.结果表明,Pol能不同程度地降低脑组织的过氧化脂质含量,提高SOD、CAT和GSH-Px活性,降低脑含水量,减轻自由基反应对脑组织的损害,对缺血的脑组织具有保护作用.Pol的作用与所用剂量有明显关系,用Pol 12mg·kg^(-1)(iv)时作用最明显,剂量过高或过低均可影响其对脑组织的保护作用.
A number of Chinese herbs have been indicated to have protective effects beforecerebral ischemic attacks in man. Polydatin, a glycoside isolated from the roots of Polygonum cuspidatum, is one of the major active ingredients of the Chinese herb. To investigate the effects of polydatin on free radicals in cerbral ischemia, a model of ischemia/reperfusion in rat brain was used. After polydatin administration, the contents of brain water and malondialdehyde (MDA) decreased, and the activities of superox-ide dismutase(SOD), catalase (CAT) and glu-tathione peroxidase (GSH-Px) were elevated to certain extent in cerebral cortex and hippocampus of the rats. Protective action of polydatin in ischemia/reperfusion induced brain injury has been suggested. The effect of polydatin was re-latd to the concentrations of the polydatin administered. The results showed that 12 mg ?kg-1 polydatin (iv) had the most significant effect on free radicals in ischemic brain injury. The results of this study may be useful in designing new drugs in the treatment of strokes and cerebral vascular diseases.
