目的:探讨肝脂酶(HL)启动子514C/T基因多态性与血脂水平的关系。方法:用聚合酶链反应-限制性内切酶片段长度多态性分析技术(PCR-RFLP),对112名三酰甘油(TG)正常水平者和103例高TG者的HL启动子514C/T基因多态性进行研究。结果:HL启动子514TT等位基因频率在高TG组为0.4563,比正常TG组0.3348高,但差异无显著性。在总研究组中TT型的年龄、TG浓度明显高于CC型和CT型,而载脂蛋白A1(ApoA1)则明显降低(P<0.05)。按性别分层后发现TT型女性和男性的TG、低密度脂蛋白胆固醇(LDL-C)的浓度均明显高于非TT型同性(P<0.05),而高密度脂蛋白胆固醇(HDL-C)、ApoA1浓度则分别低于非TT型的同性(P<0.05)。经Logistic回归分析显示总胆固醇(TC)、ApoB、TT型是高TG危险因子,而ApoA1、HDL-C则是保护因子。结论:HL启动子514C/T多态性与高TG血症的发生相关,并可影响血浆脂类的代谢,可能是高TG所致疾病的危险因素。
Objective To investigate the relationship between serum lipids and hepatie lipase (HL) promoter 514 C/T polymorphism. Methods The HL promoter 514C/T polymorphism was analyzed by polymerase chain reaction (PCR)restriction fragment length polymorphism in 112 patients with hypertriglyceridemia and 103 healthy subjects. Results The frequencies of TF were 0.4563 in the patients with hypertriglyceridemia and 0.3348 in the healthy subjects(P〉0.05 ). The age and the serum triglyceride level of TT genotype were both higher than those of CC and CT genotype (P〈0.05), while the ApoA1 level was lower than that of CC and CT genotype(P〈0.05). After the data were stratified by gender, the serum triglyceride and LDL-C levels in the female or male with TT genotype were higher than those in the corresponding gender without TT genotype, while the serum HDL-C and ApoA1 levels were lower than those in the individuals without TF genotype. The multivariate logistic regression analysis showed that the serum TC (total cholesterol), the ApoB level and the TT genotype of the HL gene were risk factors for high triglyceride level, while the ApoA1 and HDL-C levels may be the protective factors. Conclusions The HL promoter 514C/T polymorphism is associated with the hypertriglyceridemia and may regulate the metabolism of serum lipids, which might be a risk factor for hypertriglyceridemia.
