环状介导等温DNA扩增(LAMP)技术是一种新的核酸扩增方法,它能够高特异性、高效、快速地进行核酸的扩增。利用LAMP法检测乙型肝炎病毒(HBV),能够在等温条件下于1h内将少量的基因拷贝数扩增至109,在对65份临床标本的检测中显示了较高的特异性。与现有的PCR技术相比,LAMP法更加简便快速,且在等温条件下进行,不需要复杂的仪器设备,为临床检测乙肝病毒提供了一个快速简便的新方法。
Loop-mediated isothermal amplification(LAMP) is a novel nucleic acid amplification method in which reagents react under isothermal conditions with high specificity, efficiency, and rapidity. The reliability of LAMP, initially developed for the detection of HBV, was evaluated in this study. When agarose gel electrophoresis was used for the detection of LAMP products, the sensitivity of a 60 min HBV DNA LAMP reaction reached 10^9 copies/tube. Following these initial validation studies, clinical samples collected from 65 patients with primary HBV. By use of agarose gel electrophoresis, HBV LAMP products could be detected in blood samples, but no LAMP product was detectable in healthy body's blood samples. This method is rapid, taking only 2 h, compared with PCR. In addition, the LAMP method is performed under isothermal conditions and no special apparatus is needed, which makes it more economical and practical than PCR. These results indicate that LAMP can provide a rapid yet simple test for the detection of HBV.
