摘要
目的动态观察兔50%Ⅲ度烫伤早期血脑屏障(blood brain barrier,BBB)改变情况.方法雄性大白兔10只,随机分为对照组、烫伤后1,2,3,4 h 5个时相点组.烫伤组均制成50%Ⅲ度烫伤模型.在每个时相点组分别取2只兔,取脑组织行光镜及透射电镜观察.采用激光共聚焦扫描显微镜(laser confocal scanning microscope,LCSM)测量脑血管内皮细胞(vascular endothelial cell,VEC)的荧光含量. 结果病理结果显示,严重烫伤后2 h,VEC内仅有少数线粒体发生空泡变.严重烫伤后3~4 h,VEC肿胀,细胞紧密连接增宽、结构稍模糊.烫伤后2 h,VEC的荧光含量较对照组减少(P<0.05),烫伤后3,4 h较对照组有明显减少(P<0.01). 结论 LCSM通过对VEC荧光含量的定量分析,可以了解VEC的损伤程度.
Objective To explore the changes of blood brain barrier (BBB) at the early stages of severe scald in rabbits. Methods Ten male white rabbits were randomized into control, 1 hour, 2 hours, 3 hours and 4 hours groups after scald. The rabbits in scald groups were inflicted with 50% TBSA Ⅲ skin scald. Brain tissues from two rabbits from every, scald group were collected for observation under microscope and transmission electron microscope ( TEM). The fluorescence content of vascular endotbelial cells (VEC) was measured by laser confocal scanning microscope ( LCSM ) at different phases. Results The pathologic results showed that only a few mitoehondria had vacuoles two hours after scald and that VEC was swelled, with the tight junction becoming loose 3-4 hours after scald. The content of fluorescence was remarkably decreased two hours after scald ( P 〈 0.05 ) but very significantly decreased four hours after scald compared with control group ( P 〈 0.01 ). Conclusion The degree of VEC damage can be determined by LCSM quantitatively determining the fluorescence content of VEC.
