摘要
在体外用Cu^(2+)诱导法使LDL氧化修饰,利用紫外法监测氧化过程中所形成的结合二烯键,发现整个反应分为3个时相,即延迟期、快速反应期和分解期。利用硫代巴比妥反应物测定,发现氧化后LDL中每mg蛋白脂质过氧化物增加了15倍。
Low density lipoprotein(LDL)was oxidized with Cu ̄(2+)in vitro.A lag phase was followed by oxidave modification after addition of CuCl_2, and about 15 h,the reaction reach a peak. LDL oxidation was continuously monitored spectrophotometrically at 234 nm to follow the formation of conjugated dienes.Three phases were found in oxidation curve:Lag phase, propagation phase and decomposition phase. By determinating thiobarbituric acid reaction substances(TBARS),we found the lipid peroxidation increased as much as 15 times in LDL.
