摘要
静脉内注射脂多糖(LPS)6mg/kg可引起大鼠血清肿瘤坏死因子-α(tumornecrosiskctor-α,TNF-α)短时间增多和高甘油三酯血症,同时出现血压下降、心输出量减少、四肢冰凉等休克症状,动物大多于12小时内死亡。提前1小时腹腔内注射秋水仙碱(colchcine,Col.)0.5mg/kg可使LPS诱导的血清TNF-α水平升高的程度明显减轻;同时,血压维持正常,心输出量显著高于单独注射LPS组,血清甘油三酯水平正常。本文再次证实TNF-α在内毒素性休克中的重要作用,而Col.减轻休克症状的机制很可能与其抑制TNF-a的产生密切相关。
Intmvenous injection of lipopolysaccharide(LPS, 6 mg/kg)in the rat resulted in anabrupt,monophasic but tiansient elevation of the serum TNF-α level followed by a gradual in-crease in serum triglyceride co ncentration within a period o f 4 hours. Changes in these serum pamrneters were associated with shock symptoim including a dramatic fall of arterial blood pres-sure and cardiac output,cold extremities etc. majority of the animals died within 12 hours. Intraperitoneal injection of colchicine(Col., 0.5mg/kg)one hour before LPS administration brought about a marked alleviation ofthe LPS-induced elevation of serum TNF-α level. Besides, ar-terial blood pressure and serum triglyceride concertration were maintained within the normal limits in the Col. -pretreated rodents. The cardiac output,however,was apparently higher than thatin animals treated with LPS alone. The present work once more confirmed the important role of TNF-αin endotoxic shock and the mechanism underlying the relief of shock symptoms by Col. -pretreatment might be closely related to its iuhibition TNF-αproduction.
