酮康唑对卵巢过度刺激综合征模型大鼠的血管内皮生长因子及肿瘤坏死因子的影响研究

Effects of Ketoconazole on VEGF and TNF in Rat Models with Ovarian Hyperstimulation Syndrome

摘要 目的:观察酮康唑对卵巢过度刺激综合征(OHSS)模型大鼠血管内皮生长因子(VEGF)及肿瘤坏死因子(TNFα、TNFβ)的影响。方法:将OHSS模型大鼠随机分为酮康唑治疗组、白蛋白阳性对照组、阴性对照组(生理盐水)及刺激组(绒毛膜促性腺激素)。各组给予相应药物作用后切取卵巢,采用免疫组织化学染色法观察VEGF及TNFα、TNFβ在卵巢中的表达并进行比较。结果:在4组卵巢细胞胞浆内VEGF均呈阳性,治疗组及阳性对照组VEGF表达显著低于阴性对照组和刺激组(P<0.05);治疗组与阳性对照组比较无显著性差异(P>0.05)。4组中TNFα、TNFβ表达无显著性差异(P>0.01或P>0.05)。结论:酮康唑可能通过降低VEGF水平而发挥治疗OHSS的作用。 OBJECTIVE: To observe the effects of ketoconazole on VEGF and TNFα, TNFβ in rat models with ovarian hyperstimulation syndrome(OHSS). METHODS: The rat models with OHSS were randomly divided into ketoconazole treatment group, albumin positive group, negfitive control group (saline), and stimuli group( camisia foetus gonad promoting hormone). Each group was administered with relevant medication, and then the ovary was removed,expressions of VEGF and TNFα,TNFβ in ovary were observed by immunohistological stain and compared among 4 groups. RESULTS:Positive immunostain of VEGF was located in cytoplasm of all 4 groups, the expression of VEGF was significantly lower in treatment and positive control group than that in negative control and stimuli group(P〈0.05) . No significant difference was found between the treatment and positive control group(P 〉 0.05) . There was no significant difference in the expressions of TNFa and TNFα among 4 groups(P〉0.01 or P〉 0.05).. CONCLUSION: Ketoconazole may play its role in treating OHSS by reducing the expression of VEGF .
