摘要
研制了盐酸拉贝洛尔缓释片,研究了其人体内药物动力学。缓释片体外可维持释药12h,释药行为可用Higuchi方程来描述,溶出速度常数为4.26min ̄(-1/2);其体内药动学过程符合一级吸收单室模型,药动学参数为:Ka=1.71h ̄(-1),K=0.1248h ̄(-1),Cmax=61.93ng/ml,Tmax=1.89h,AUC ̄(0→∞)=603.9ng·h/ml。
Sustained release tablets(SRT)of labetalol and the human pharmacokinetics were studied. The results indicated that the SRT was able to produce a sustained release of the drug for 12 h.The drug release pattern in vitrO of the SRT was best described by Higuchi equation with a release constant being 4.26 min and the plasma drug level data best fit the one compartment open model with first order absorption. The parmacokinetic parameters Kα,K,C(max),T(max) and AUC of labetalol HCl were 1.17 h-1,0.1248 h-1, 61.93 ng/ml,1.89 h and 603.9 ng·h/ml respectively.
