大蒜素对脂多糖致急性肺损伤小鼠防治作用的实验研究 被引量:10

摘要 目的探讨大蒜素对脂多糖(LPS)致急性肺损伤(ALI)小鼠的防治作用。方法复制LPS致ALI小鼠模型。实验动物随机分为生理盐水对照组,ALI组,大蒜素预防组,大蒜素治疗组,大蒜素组。测定各组26、h肺湿重/干重比值(W/D)。用酶联免疫吸附法(ELISA)测定各组2、6 h血清和肺组织匀浆上清中肿瘤坏死因子-α(TNF-α)及白细胞介素-1β(IL-1β)浓度。观察各组肺组织病理改变。结果ALI组,2 h肺W/D较对照组无明显变化,6 h肺W/D显著高于对照组(P<0.05)。而大蒜素预防组可使肺W/D显著降低(P<0.05),但在大蒜素治疗组肺W/D无明显改善。大蒜素组较生理盐水对照组无明显变化。肺组织病理显示,在ALI组12 h时可见肺间质充血、水肿,大量炎性细胞浸润,大蒜素预防组可明显改善肺组织病理变化,大蒜素治疗组无明显变化。ALI组,2 h血清及肺组织中TNF-α及IL-1β含量均明显升高(P<0.01),6 h有所降低,但仍高于对照组(P<0.01);大蒜素预防组可明显降低血清及肺组织中TNF-α及IL-1β的含量,但大蒜素治疗组无明显效果。结论预先给予大蒜素可明显减轻LPS诱导的ALI小鼠肺组织炎症反应及肺水肿程度,对血清及肺组织中TNF-α及IL-1β的生成具有抑制作用。 Objective To investigate the effect of allicin on mice with acute lung injury (ALI) induced by lipopolysaccharide (LPS). Methods ALI murine models were duplicated by injection of LPS intraperitoneally. Mice were randomly divided into saline control group, ALl group, allicin prevention group, allicin treatment group, and allicin group. The ratio of pulmonary wet weight to dry weight (W/D) was measured to assess the extent of pulmonary edema. Lung pathological changes were also observed in each group. Tumor necrosis factor-α (TNF-α) and interleukin-1β(IL-1β) levels in the serum and the supernatant of lung homogenates were detected by ELISA. Results At 2h of ALl group, no change was detected in lung W/D compared with control group. At 6h of ALl group, the lung W/D was significantly elevated(P〈0.05), and it was reduced in allicin prevention group, but no decrease was observed in the allicin treatment group. Histologically, massive alveolar interstitial tissue edema, hemorrhage, and inflammatory cell infiltration were observed at 12h of ALI group, and they could be alleviated in allicin prevention group. The level of TNF - α and IL-1β in serum and supernatant of lung homogenates increased markedly at 2h of ALI group(P〈0.01), and decreased at 6h, which were still higher than those in the control group(P〈0.01). The production of TNF - α and IL - 1β induced by LPS could be reduced in allicin prevention group, but not be found in allicin treatment group. Conclusions Preliminary administration of allicin alleviates the degree of pulmonary edema and pulmonary inflammation, and inhibits the production of TNF - α and IL - 1β in mice with acute lung injury induced by LPS.
