菊花叶片不定芽再生体系的研究 被引量:30

摘要 以7个不同菊花品种的叶片为外植体,探讨了基因型、苗龄、叶片着生部位、Vc、活性炭、AgNO3以及不同生长调节剂组合等因素对菊花叶片不定芽再生的影响.试验结果表明,基因型是影响不定芽再生的重要因素;15~35d苗龄的无菌苗中、上部幼嫩叶片不定芽再生能力较高;高浓度NAA有利于提高‘紫妍’叶片外植体的再生能力和消除褐化现象;AgNO3未能促进叶片外植体的再生;适宜‘紫妍’再生的培养基是MS+6BA1mgL+NAA0.5mgL和MS+6BA2mgL+NAA1mgL;适宜‘L12M57’再生的培养基是MS+6BA2mgL+NAA0.2mgL和MS+6BA2mgL+NAA0.1mgL. The effects of different factors, such as genotype, seedling age, position of leaves on seedling, Vc, charcoal, AgNO3 and different hormone combinations, on shoot regeneration from leaves of seven different Dendranthema×grandiflora cuhivars were studied. Results indicate that the genotype is an important factor in shoot regeneration. The infant leaves from seedlings with an age of 15-35 d have high shoot regeneration. High concentrations of NAA accelerate shoot regeneration of Dendranthema×grandiflora ‘Ziyan' . No positive effect of AgNO3 on shoot regeneration is observed. The optimum medium for ‘Ziyan' is MS + 6-BA 1 mg/L + NAA 0.5 mg/L and MS + 6-BA 2 mg/L+ NAA 1 mg/L.MS + 6-BA 2 mg/L+ NAA 0.2 mg/L and MS + 6BA 2 mg/ L + NAA 0.1 mg/L is suitable for‘ L12M57' .
