环氧合酶-2抑制剂尼美舒利对肝纤维化大鼠胃黏膜损伤的影响 被引量:1

Effects of cyclooxygenase-2 inhibitors Nimesulide on healing of gastric mucosal damage in fibrotic rats

摘要 目的研究探讨COX-2抑制剂尼美舒利对CCL4诱导的肝纤维化大鼠胃黏膜损伤愈合的影响。方法取雄性SD大鼠20只,按体重随机分为两组,模型组(n=10):40%CCL4油0.3mL/100g体重灌胃,每周2次;尼美舒利治疗组(n=10):40%CCL4油0.3mL/100g体重灌胃,每周2次,同时给予尼美舒利6mg/(kg·d),每天1次,于第8周取材,光镜下观察肝脏组织学改变,进行胃黏膜损伤和炎症指数评价。结果尼美舒利治疗组大鼠肝组织纤维化程度减轻,纤维间隔较为细小,且胃黏膜炎症评分明显低于模型组(P<0.01),损伤评分亦低于模型组(P<0.05)。结论尼美舒利能够在一定程度上减轻肝纤维化大鼠的肝纤维化程度,对其损伤的胃黏膜有保护作用,促进愈合。 [Objective] Cyclooxygenase(COX),the key enxyne for synthesis of prostaglandins (PGs), exists in two isoforms (COX-1 and COX-2). Conventional nonsteroidal anti-inflammtory drugs (NSAIDs) are limited in application as it inhibitor both COX-1 and COX-2 activities simultaneously and induce serious gastrointestinal side effects. But, specific COX-2 inhibitors are expected to become a new type of NSAIDs without much gastric side effects because it can inhibit COX-2 activity, only. The aim of this study was to investigate the effects of cyclooxygenase-2 inhibitors Nimesulide on the healing of gastric mucosal damage in fribrotic rats induced by CCIA. [Methods] 40 male wistar rats were randomly divided into 2 groups in term of weight. Model group (n =10):the rats were treated with 40% CCIA 0.3 mL/100 g body weight by daily gastrogavage two times a week. Nimesulide group(n =10): the rats were treated with 40% CCIA 0.3 mL/100 g body weight by daily gastrogavage two times a week. Nimesulide was given 6 mg/(kg, d) only one time a week at the same time. After 8 weeks, liver morphological examination was based on microscopy. Meanwhile, the score of gastric mucosal impairment and inflammtion were assessed. [Results] Mean fibrosis was reduced in Nimesulde treatment group, and there was a marked decline of both inflammatory score (P 〈 0.01 ) and injured score (P 〈0.05) in it, compared to those of the model group. [Conclusions] Nimesulide can not only alleviate fibrosis on fibrotic rats to some extent, but also offer protective effects on injured gastric mucosa and accelerate their healing.
