摘要
[目的]观察桂枝乙醇提取物对大鼠离体胸主动脉环的舒张作用及其可能的机制.[方法]采用大鼠离体主动脉环灌流模型,观察桂枝乙醇提取物对苯肾上腺素(1.0μmoL/L)预收缩内皮完整的血管环和去内皮血管环的舒张作用,并观察一氧化氮合酶抑制剂L-硝基精氨酸甲酯和鸟苷酸环化酶抑制剂亚甲蓝预处理对桂枝乙醇提取物舒张血管作用的影响.[结果]桂枝乙醇提取物对离体大鼠胸主动脉环具有非内皮依赖性的舒张作用,分别用L-硝基精氨酸甲酯和亚甲蓝预处理对桂枝乙醇提取物的舒张血管作用无明显影响.在无钙缓冲液中,桂枝乙醇提取物对血管收缩有抑制作用.[结论]桂枝乙醇提取物对大鼠离体胸主动脉环的舒张血管作用具有非内皮依赖性,其机制可能与抑制血管平滑肌细胞内质网储存钙的释放有关.
OBJECTIVE To study the vasodilative effects and its possible mechanisms of the ethanol extract of Ramulus Cinnamomi(RC) on thoracic aorta ring of rats in vitro.METHODSIt was observed by using the perfusion model of thoracic aorta ring of rats in vitro that the vasodilative effects of the ethanol extract of RC for endothelium-intact or endothelium-denuded thoracic aortic rings pre-contracted by phenylephrine(PE,1.0μmoL/L) and pre-treated by N-nitro-L-arginine methyl ester(L-NEME) of nitric-oxide synthase(NOS) inhibitors and methylen blue(MB) of guanylate cyclase inhibitors.RESULTSThe ethanol extracts of RC had endothelium-independent vasodilative effects for thoracic aorta ring of rats in vitro by PE,no vasodilative effects by L-NEME and MB,and inhibited vasoconstrictive effects in the buffer solution without extracellular calcium.CONCLUSIONThe ethanol extracts of RC has endothelium-independent vasodilative effects for thoracic aorta ring of rats in vitro,and its mechanisms maybe is correlated with the inhibition of calcium release stored in the endocytoplasmic reticulum of vascular smooth muscle cell.
