蛋白酶体抑制剂MG-132增强4-羟苯基维胺脂诱导的肺癌细胞凋亡

摘要 [目的]观察4-羟苯基维胺脂(4-HPR)联合应用蛋白酶体抑制剂MG-132对肺癌A549细胞凋亡的影响.[方法]用倒置显微镜和TUNEL染色观察4-HPR或(和)MG-132联合应用后A549细胞形态学变化及对细胞生长的抑制作用;免疫印迹法检测核转录因子-κB(NF-κB)的表达.[结果]不同给药组细胞数量明显变少,失去正常的生长,细胞密度低,光泽度下降,肿胀变形,变圆;4-HPR加MG-132组细胞凋亡率明显升高,MG-132明显增强4-HPR诱导的A549细胞凋亡;免疫印迹观察显示,MG-132降低4-HPR诱导的NF-κB的表达.[结论]MG-132是通过降低NF-κB的表达而增强4-HPR诱导的A549细胞凋亡. OBJECTIVE To study the combined effects of 4-HPR and MG-132 induced apoptosis in lung cancer A549 cells.METHODS The cellular morphology and inhibition growth efficacy on A549 cell was assessed by inverted microscope and TUNEL,and the expression of NF-κB was examined by Western blot.RESULTS The MTT analysis indicated MG-132 could obviously enhanced 4-HPR-induced apoptotic cell death in A549 cells,subsequent observations using Western blot indicated that MG-132 effectively inhibits NF-κB activation during 4-HPR-induced apoptosis.CONCLUSION MG-132 can enhance 4-HPR-induced A549 cells apoptosis via inhibition of NF-κB activiation.
