摘要
使用2.1μm的单分散无孔硅胶作为基体,表面化学键合C_8和C_(18)非极性固定相,并在这种键合相和相关的大孔硅胶上考察了蛋白质分离的色谱性能。实验结果表明,梯度冲洗下该固定相能使核糖核酸酶、胰岛素、溶菌酶、牛血清蛋白和卵清蛋白得到好的分离,分析速度快、效率高,为生物大分子的高效液相色谱分离开辟了另一条途径。
n this work,the bonding of C_8 and C_(18) to 2.1μm monodisperse non-porous silica for the preparation ofnovel hydrophobic packings has been studied.The chromatographic properties of the non-porous packingsand large pore packings were examined and compared.The results showed that good revesed-phase bondedsilica packings can be prepared.Under gradient elution. proteins(RNase,Ins,Lys,BSA and Ova)wereseparated within 15min successfully. It is suggested that the non-porous packings are a better alternative forthe biopolymer separation.
