摘要
研究了侧耳属白腐真菌Pleurotssp.在木屑、玉米秆和稻草秆三种固体秸秆基质上分泌漆酶活性、漆酶动力学参数米氏常数Km和漆酶同工酶的动态变化.结果表明:该菌株在不同秸秆基质上分泌漆酶规律不一致;漆酶Km值随时间变化很大,相差高达一个数量级;通过PAGE活性电泳研究漆酶性质,表明体系中存在多种漆酶同工酶,不同时间不同秸秆基质上漆酶同工酶数量和性质存在很大差异,木屑固体秸秆基质上漆酶同工酶Ⅰ的表达比另外两种固体秸秆基质更明显,同时还发现漆酶同工酶谱带和酶活性大小存在一定的关系,基本上漆酶活性越高,同工酶谱带Ⅰ颜色就越深越长,而同工酶Ⅱ和Ⅲ只起辅助作用.
The dynamic process of laccase produced by white rot fungus Pleurots sp. , kinetics constant Km of crude laccase and laccase-isoform during the culture of wood scraps, corn stalks and straw stalks the three solid states was studied. There are different regulations of laccase in different media. The range of change of Km can even exceed by 10 times. Laccase-isoforms were detected through active-PAGE electrophoresis,showing that there is much difference in their amont and characters when the media and time are different.There are some relations between the laccase-isoform bands and lacccase activity. The deeper and the longer the band of Lac I is, the higher the activity is. Lac Ⅱ and Lac Ⅲ just play a supplementary role.
