摘要
①目的观察甲壳质衍生物壳寡糖对骨折愈合过程中成骨细胞内骨形态发生蛋白(BMP)的影响。②方法健康新西兰兔40只,制备右桡骨中段3mm骨缺损模型,随机分成对照组和实验组,分别灌胃给予生理盐水和壳寡糖。于术后9、17、30、42d,苏木精伊红染色观察骨折愈合情况,免疫组化方法观察骨折局部BMP蛋白的表达。③结果苏木精伊红染色显示实验组术后的骨折愈合情况比对照组好;免疫组化结果显示术后9、17d时,实验组骨折局部BMP含量均较对照组明显增高,差异有显著性(t=15.579、28.817,P<0.01);实验组BMP高峰出现在术后17d,而对照组出现在术后30d。④结论壳寡糖能促进成骨细胞合成BMP,使BMP峰值提前,从而加速骨折愈合。
Objective To study the effect of chitowoligosaccharide on BMP content in osteoblast in the process of fracture healing. Methods Forty healthy New Zealand rabbits were made into models with 3 mm bone defect in the middle segment of radius, and randomly divided into the control group and experimental group. They were respectively given normal saline and chitowoligosaccharide daily through gastric tube. HE staining was used to observe the healing and immunohistochemistry was applied to observe the expression of BMP proteins at 9,17,30, and 42 days postoperatively. Results HE stain showed that the fractures healed better in the experimental group than the control. The expression of BMP in the fractures in the experimental group at 9 and 17 days was significantly higher compared with the control group (t=15. 579,28. 817; P〈0.01). The highest BMP content appeared at 17 days in the experimental group, while at 30 days for the control one. Conclusion Chitowoligosaccharide could promote osteoblast to synthesize BMP and the expression of BMP to accelerate fracture healing.
