肉鸡骨髓造血祖细胞体外培养及增殖功能研究 被引量:1

STUDIES ON IN VITRO PROLIFERATION OF HEMATOPOIETIC PROGENITOR CELLS IN THE CHICK

摘要 对肉鸡骨髓红系和粒单系祖细胞集落形成单位(CFU-E和CFU—GM的培养条件和生长情况进行了探讨,并观察了不同日龄肉鸡骨髓造血祖细胞体外增殖能力。结果表明:(1)培养体系中加入灭活鸡血清,其集落产率较加小牛血清为高,其生长分裂也迅速,说明同源血清优于异源血清。(2)肉鸡骨髓CFU—E培养体系最适pH值为7.8~8,0;CFU—GM最适pH值为7.2~7.3。(3)试验使用鸡脾细胞条件培养液作为集落刺激因子,CFU—GM生长良好。(4)7日龄肉鸡CFU—F为153±8.15,CFU—GM为162±3.94.42日龄肉鸡CFU—E为62±2.12,CFU—GM为83±3.24。两种日龄红系、粒系集落产率均有明显差异(P<0.01),表明肉鸡骨髓造血祖细胞功能状态不同,呈现日龄差异。 Culture methods for the in vitro development of bone marrow (B M) CFU E and CFU-GM in broilers, were studied and the colony number was also compared between 7 day-old and 42 day-old chicks. The results were as follwos.1. The number of colonies obtained with culture medium containing chicken serun was higher than that obtained with the medium containing bovine serum. 2. The Optimal pH for the development of broiler CFU-E and CFU-GM was 7.8 ̄8.0and 7.2 ̄7.3, respectively.3. The CFU ̄GM growed well when the chicken spleen-conditional medium was used as a colony-stimulating factor. 4. The plating efficiencies of BM CFU-E were 153±8, 15 and 63±2.12/5×105, andthose of CFU-GM were 162±3.94 and 83±3.24/5×105 at the age of 7 days and 42 dayts, respectively. The differenge in the plating effieieney between the two ages was signinficant (P<0.01), and this indicated a different proliferative potential in BM hematopoietic progenitor cells at the different ages of broilers.
