摘要
目的建立HPLCMS同时检测6种马兜铃酸类成分的方法。方法采用高效液相质谱联用法检测,色谱柱为DiamonsilC18柱(250mm×4.6mm,5μm);乙腈水系统,分别含0.1%的乙酸和0.1%的四氢呋喃,采用梯度洗脱程序;流速:1.0ml/min,采用DAD检测,波长范围190~400nm。质谱采用ESI负离子模式和选择性离子扫描(SIM)。结果马兜铃酸A、B、C、D、7OH马兜铃酸A和去硝基马兜铃酸A等6种成分可在1h内达到分离并得到同时检测。结论该法可用于含有或可疑含有马兜铃酸类有毒成分中药材及制剂的检测,方法灵敏,快速,专属性较强。
Aim To determine six constituents in the fruits of Aristtolochia contorta.Methods The HPLC-UV-MS analysis was carried out on Diamonsil C(18) column (250mm×4.6mm,5μm),the mixture of acetonitrile-water was used as the mobile phase by a gradient program,which contain 0.1% acetic acid and 0.1% tetrahydrofuran.The flow rate was 1.0ml/min.Results A high-performance liquid chromatographic method combined with a photodiode array detector (HPLC/UV) and mass spectrometry (LC/MS) for analysis of aristolochic acid A,B,C,D,7-OH-aristolochic acid A,and aristolic acid in a number of plant materials and their commercial products was developed.Conclusion HPLC with DAD detector and ESI mass spectrometry experiments allowed the identification of the target compounds and increased the selectivity of complex analyses such as those involved with multi-botanical preparations.
