免疫胶体金与酶免疫检测HIV抗体和梅毒螺旋体抗体对照实验 被引量:12

Controlled experiments of testing anti-TP and anti-HIV by DIGFA and EIA

摘要 目的采用免疫胶体金技术(DIGFA)检测试卡和酶免疫(EIA)检测试剂,检测艾滋病病毒抗体(抗HIV1/2)和梅毒螺旋体特异性抗体(抗TP),对DIGFA技术参数进行评估。方法对多份质控血清和5863份患者血清或血浆,分别采用DIGFA试卡和EIA试剂进行检测,以EIA检测结果为参照,对DIGFA检测试卡的灵敏度、特异性、检测效能和物理性能等参数进行评价。结果多份质控血清抗TP和抗HIV1/2DIGFA试卡检测特异性均为100%;抗TP、抗HIV1/2DIGFA试卡检测的灵敏度分别为80.00%和93.33%;检测效率分别为88.44%和96.97%。5863份患者血清(血浆)的抗TP和抗HIV1/2DIGFA试卡检测特异性分别为99.86%和99.76%;灵敏度分别为50.94%和77.78%;检测效率分别为99.42%和99.69%。结论DIGFA试卡检测灵敏度比较低,花费成本较大,不适宜献血员筛选试验。如果应用到街头(采血车)献血员快速筛选,必须和EIA联合应用。 Objective To evaluate the quality and performance of 3 sources of Dot-immuno gold filtration assay(DIGFA) calories and EIA kits for testing anti-TP and anti-HIV1/2,and to provide data for various laboratories of blood centers in selecting anti-TP and anti-HIV1/2 assay methods.Methods Several known quality control blood sera and 5 863 unknown blood sera or blood plasmas of patients were tested by DIGFA calories and EIA.The sensitivity,specificity,physical properties and consistency of the calories were assessed.Results In several known quality control blood sera,the specificity of anti-TP and anti-HIV1/2 were found to be 100%,the sensitivity of anti-TP and anti-HIV1/2 was 80.00% and 93.33% respectively,and the efficiency of anti-TP and anti-HIV1/2 was 88.44% and 96.97% respectively.In 5 863 unknown blood sera or blood plasmas of patients,the specificity,sensitivity and efficiency of anti-TP and anti-HIV1/2 were found to be 99.86% and 99.76%,50.94% and 77.78%,99.42% and 99.69%.Conclusion DIGFA is less sensitive than EIA and more expensive.It is fit for emergency rather than for rapid screening of blood donors.In blood donors's rapid screening,DIGFA should be combined with EIA,since screening by DIGFA alone may not be reliable.
