兔动脉粥样硬化模型的建立及丹皮酚药物作用的实验研究 被引量:29

ESTABLISHMENT OF ATHEROSCLEROTIC MODEL IN RABBITS AND EXPERIMENTAL STUDY ON PAEONOL ACTION

摘要 目的探讨兔动脉粥样硬化模型的建立方法及评价标准,观察丹皮酚对模型动物的治疗作用。方法通过高脂饲料喂养12周建立兔动脉粥样硬化模型,将模型兔随机分为3组,丹皮酚小剂量组、丹皮酚大剂量组和模型对照组,每组各6只,灌胃给药6周,模型和空白对照组给予同体积的0.5%CMC-Na。6周末测定血清脂质,光镜下观察各组动物主动脉组织形态学改变。结果高脂饲料喂养家兔12周可形成稳定的兔动脉粥样硬化模型。模型组动物血清TC和LDL显著升高,丹皮酚大、小剂量组可明显降低血清TC和LDL。病理学观察显示模型组可见典型的粥样斑块形成,而丹皮酚用药组可不同程度地减轻主动脉病变。结论高脂饲料喂养法形成的兔动脉粥样硬化模型方便可靠,口服丹皮酚可延缓动脉粥样硬化进展、改善主动脉组织病变程度。 Objective:To evaluate the establishment of atherosclerotic model in rabbits and observe the therapic effects of paeonol on it.Method:The rabbits were given high cholesterol diet for 12 weeks.After the model had been established,the rabbits fed with high cholesterol diet were divided into 3 groups:paeonol low dose,paeonol high dose and model group.The paeonol groups were administered paeonol once a day for 6 weeks,while the model and normal group were given the same volume 0.5% CMC-Na solution.After 6 weeks treated,the level of serum total cholesterol(TC)?low-density lipoprotein cholesterol(LDL-C)were determined with biochemical techniques and the pathomorphological changes of rabbits'aortae was observed with pathological techniques.Results:The rabbits fed with high cholesterol diet for 12 weeks have formed stable atherosclerosis.The values of TC and LDL in model group were obviously raised,while it was markedly decreased in paeonol groups.A typical pathological manifestation of atherosclerosis was founded in model group.The groups fed with paeonol expressed a less serious lesions of aortic intima than that of the model.Conclusion:The method of establishing atherosclerotic model in rabbits fed with high cholesterol diet is reliable.The paeonol can effectively suppress the development of the atherosclerotic lesion and attenuate the aortic lesions of atherosclerotic rabbits.
