摘要
目的:探讨扶正活血方对慢性肾功能衰竭(CRF)大鼠肾组织纤维连接蛋白(Fn)和转化生长因子β1(TGF-β1)mRNA表达的影响。方法:采用5/6肾切除制作大鼠 CRF 模型。模型制作成功后随机将模型大鼠分为模型组、扶正活血方组、黄芪组、丹参组、依那普利组,并与正常组对照。治疗2个月后,观察各组 CRF 大鼠肾组织 Fn 和 TGF-β1 mRNA 表达情况。结果:模型组血肌酐(SCr)、尿素氮(BUN)、24小时尿蛋白定量(24hpro)明显高于正常组,肾组织 Fn 和 TGF-β1 mRNA 表达明显强于正常组。经过扶正活血方治疗后,肾组织 Fn 和 TGF-β1 mRNA 表达明显减弱,蛋白尿水平明显降低。结论:扶正活血方可改善CRF 大鼠肾功能,延缓 CRF 进展,其机制可能与降低 CRF 大鼠肾组织 Fn 和 TGF-β1 mRNA 表达有关。
Objective:To probe effects of Fuzheng Huoxue Recipe on expression of fibronectin(Fn)and transforming growth factor-β1 mRNA (TGF-β1 mRNA)in the renal tissue of the rat with chronic renal failure(CRF).Methods:CRF rat model was established by 5/6 nephreetomy,and then they were randomly divided into model group,Fuzheng Huoxue Decoction group,Huangqi group,Danshen group and Enalapril group,with a normal control group.After treatment of 2 months,expression of Fn and TGF-β1 mRNA in the renal tissue of the rat with CRF were observed.Results:Blood creatinine(SCr),urea nitrogen(BUN),24-hour protein in urine and expression of Fn and TGF-β1 mRNA in the renal tissue in the model group were significantly higher than those in the normal control group.After treatment with Fuzheng Huoxue Recipe,expression of Fn and TGF-β1 mRNA in the renal tissue and protein level in urine decreased significantly.Conclusion:Fuzheng Huoxue Recipe can improve renal function of the rat with CRF,and delay development of CRF,which are possibly related with the decrease of expression of Fn and TGF-β1 mRNA in the renal tissue of the rat with CRF.
