摘要
目的 观察泰索帝合并吐温80对人乳腺癌MCF 7细胞增殖及PCNA表达的影响。方法 应用MTT法、免疫细胞化学和Western 印迹等技术,检测MCF -7细胞经不同浓度的泰索帝合并吐温 80作用后不同时间(2 4、4 8h)增殖活性和PCNA表达的改变。结果 泰索帝能抑制MCF- 7细胞增殖,抑制效应随着药物剂量的增加和作用时间的延长而增强,吐温80增强泰索帝的抑制效应。PCNA在MCF- 7细胞为强阳性,主要分布于细胞核;吐温组表达略有减少;泰索帝作用亦使PCNA的表达降低,且随泰索帝剂量的增加、作用时间的延长抑制愈明显;泰索帝与吐温 80合并作用后,对MCF- 7细胞PCNA表达的抑制作用较泰索帝及吐温80单独作用显著增强。结论 泰索帝合并吐温 80可显著抑制人乳腺癌MCF- 7细胞的增殖,两者存在协同效应。
Objective To study the influence of Docetaxel c ombined with Tween 80 on the expression of PCNA and cell proliferation in human breast cancer cell line MCF-7.Methods Different dose Docetaxel in combination with Twee n 80 was administered to MCF-7 cells.The expression of PCNA was assayed by immu no-cytochemistry and Western-blotting method.The cell proliferation inhibition ratio was measured by MTT assay. Results Docetaxel could inhibit the proliferation of MCF -7 cells,which was enhanced with the increasing of Docetaxel dose and treatment time.Tween80 could enhance the cytotoxicity of Docetaxel.In combination with Tw een 80,Docetaxel could dramatically suppress the expression of PCNA in a dose-d ependent and time-dependent way.Significant difference of PCNA expression was o bserved in comparison with that in Docetaxel or Tween 80 treatment alone. Conclusion The results demonstrated that Docetaxel combin ed with Tween 80 can significantly inhibit the expression of PCNA,which might im ply the blocking of proliferation of MCF-7 cells.
