摘要
一株羊轮状病毒经初生小牛肾原代细胞传代，定名为LLR－85株。按Jennerian种痘法的原理，取第37代和第19代毒种，作为人用口服轮状病毒活疫苗的候选疫苗株。疫苗株电镜检查形态典型，抗原鉴定为A群、亚群Ⅰ、血清型G10和P12。电泳型为长型，11条带。病毒滴度可达105.0～7.0TCIDEA50。经无菌试验、支原体检查、外源因子检查及动物安全试验等项目检定，均符合我国规程要求。疫苗株遗传性状稳定，有望用于婴幼儿轮状病毒急性腹泻的预防。
A strain of lamb rotavirus was passaged in primary kidney cells of newborn calf and termed LLR- 85. Based on the Jennerlan approach, the 37th and 19th passages of the strain were developed as candidates for oral rotavirus live vaccive. Electron microscopic observation showed that the candidates were normal in shape. Antigenic characteristics detection proved that the candidates were groupA, subgroup Ⅰ, serotype G10 and P12, and long eleetropherotype. The virus tlters of the candidates reached 105.0 ~107.0TCIDEA50. All the results of sterility test, mycoplasma examination. extraneous agents examination and animal sa ty test could meet the Chinese Requirements for the Biological Products. The candidates showed stable inherited charasters. thus might to be used for prevention of acute diarrhea caused by rotovirus in infants.
