丙烯腈对体外培养大鼠胚胎脊髓神经细胞增殖分化的影响

摘要 目的研究丙烯腈(ACN)对体外培养的大鼠胚胎脊髓神经细胞增殖分化的影响。方法16d的大鼠胚胎脊髓组织作原代细胞培养,从细胞形态学及细胞计数观察研究细胞生长与分化过程,同时测定蛋白质含量和丙二醛(MDA)含量并与对照组进行比较。结果10.0~100.0mg/LACN组明显抑制脊髓神经细胞分化、增殖、集落形成率明显减少,细胞体积小,细胞内MDA含量增加,蛋白质相对含量降低,并显示浓度效应关系。结论ACN引起的大鼠胚胎脊髓神经细胞的增殖和分化抑制,可能与ACN诱发蛋白质合成下降和脂质过氧化物有关。 Objective To explore the effects of acrylonitrile (ACN) on proliferation and differentiation of spinal cord nerve cells of rat embryo in vitro. Methods The primary culture of spinal cord nerve cells of rat embryo at the age of 16 days was used. The cultured cells were assessed by cellular morphological markers and cell counting. Concentration of 50 percent (IC_~50 ) inhibition were calculated and the content of protein and malondialdehyde (MDA) were determined.Results The decrease of the numbers of cells and colonies was shown in all cells treated by ACN. The contents of protein were concentration-dependently decreased and levels of MDA were increased with increasing dosage of ACN.Conclusion The inhibition of differentiation and proliferation of the embryo spinal cord nerve cells might be associated with the decrease of protein synthetics and lipid peroxidation induced by acrylonitrile.
