摘要
目的:考察羟喜树碱脂质体的粒径对组织分布的影响。方法:小鼠尾静脉注射不同粒径的羟喜树碱脂质体,采用HPLC法测定不同时间点小鼠组织脏器中的药物浓度。结果:减小粒径可延长羟喜树碱脂质体在血液中滞留的时间。不同粒径(2 5nm~2 73nm)的羟喜树碱脂质体对各组织脏器的趋向性一致,主要分布于肝脏、脾脏、肾脏和肠。结论:粒径(在2 5nm~2 73nm范围内)没有改变脂质体的组织分布特异性,但羟喜树碱脂质体在一些脏器的经时分布发生了变化。
AIM:To study the influence of particle size on th e tissue dist ribution of hydroxycamptothecin(HCPT) liposomes.METHOD:The mice w ere injected various sizes (273 nm±73 nm,153 nm±33 nm,25 nm±6 nm) of HCPT l iposomes through tail vain.The HPLC-UV met hod was used to determine the drug concentration of various tissues.RES ULT:The retention time of HCPT liposomes in blood was prolonged when the l iposome size was decreased and HCPT liposomes with various particle sizes had th e similar tendency towards the tissues in mice,i.e.,mainly targeted to liver,sp leen,kidney and intestine. CONCLUSION: Particle size didn′t chan ge the tissue specificity of HCPT liposomes in the tissue distribution, while the drug c oncentration curve versus time was altered.
