摘要
本文试验了微甘菊愈伤组织诱导的影响因素 ,结果表明 :芽尖和嫩茎作为外植体时诱导率较高 ;最适诱导培养基是MS培养基附加激素 2 ,4 -D2 .0mg/L +KTl.0mg/L ,pH值 5 .7;于 2 5± 1℃条件下黑暗诱导。另外优化了微甘菊愈伤组织继代培养条件 :浓度为 5 .0mg·L-1 的抗坏血酸 (Vc)对于微甘菊愈伤组织褐变情况有良好的抑制作用 ;继代培养基中较好的激素组合为 (2 ,4 -D1 .0mg·L-1 +NAA2 .0mg .·L-1 +KT0 .5mg .·L-1 +BA0 .5mg .L-1 ;添加 1 5 %新鲜椰子汁能够明显促进愈伤组织生长 ,而且对褐变有一定的抑制效果。依此反复继代的微甘菊愈伤组织有良好的增殖效果。
Factors affecting induction of callus in Mikania micrantha Kunth was studied in the paper. The result showed that auxiliary shoot and bud tip has a higher level of callus induction ratio: the best callus induction medium was MS wi th the addition of 2,4-D2.0mg/L+KT1.0mg/L, pH 5.7;culture in dark under 25±1℃。While callus subculture conditions has been optimized , that is cultured with the addition of coconut juice 15%, Vc 5.0mg·L -1,2,4-D0.5mg/L+NAA2.0mg/l+KT0.5mg/L+BA0.5mg/L,and the callus of Mikania micrantha Kunth could inhibit browning effectively and good growth effect after continuous subculture.
