摘要
用反相 HPLC 测定了家兔口服吲哚美辛锌胶囊和混悬剂后血浆中吲哚美辛的含量,其药时曲线均符合一室开放模型,药动学参数分别为:Cmax2.1、2.4μg/ml;Tmax5.2、4.9h;k_e0.12、0.13h^(-1);k_a0.29、0.30h^(-1);t_(1/2)5.6,5.5h;t_(1/2a)4,2.3h;AUC31.9、36.1μg·h/ml。胶囊剂相对于混悬剂的生物利用度为88.9%
A HPLC method has been used for the determination of indome-thacin in plasma.After single dose of zinc-indomethacin capsules and suspen-sion in rabbits,plasma concentration-time curves corresponded to one compar-tment open model.The pharmacokinetic parameters of zinc-indomethacin capsulesand suspension were:C_(max)=2.1 and 2.4μg/ml;T_(max)=5.2 and 4.9h;k_a=0.29 and0.30h^(-1);k_e=0.12 and 0.13h^(-1);t(1/2a)=2.4 and 2.3h;t_(1/2)=5.6 and 5.5h;AUC=31.9and 36.1μg.h/ml,respectively.The bioavailability of the capsules relative tothe suspension was 88.9%.
