大骨节病区FA对培养软骨细胞分化的影响 被引量:2

摘要 目的为探讨大骨节病区腐植酸(FulvicAcid,FA)对培养软骨细胞分化的影响。方法采用SDS-聚丙烯酰胺凝胶电泳和West-ern免疫印迹及Northern杂交法检测FA作用于体外长期培养的软骨细胞;采用荧光法检测碱性磷酸酶(AlkalinePhosphatase,AL-Pase)活性。结果发现Ⅰ型胶原蛋白产生及其mRNA表达,在一定浓度范围内与FA浓度是正相关,但是当FA浓度大于50mg/L时细胞出现坏死,Ⅰ型胶原蛋白产生量降低;发现ALPase活性在一定浓度范围内,随FA浓度增大也增强,但当FA浓度增大到50mg/L时,ALPase活性降低。结论提示FA对大骨节病患者软骨细胞有加速分化作用,可能与大骨节病病理过程相关。 To explore the effects of fulvic acid (FA) from Kashin-Beck Disease (KBD)area on differentiation of chondrocytes. Methods Human embryo chondrocytes were exposed to different concentrations of fulvic acid. The content of type I collagen was detected by SDS-PAGE and the activity of alkaline phosphatase (ALPase) was detected using the fluorometric substrate, 4-methylumbelliferyl phosphate. Results In certain concentration range,with rising of FA,the content of type I collagen increased (<50mg/L ),but when exposed to high concentration (>50mg/L),cells were damaged and secreted less type I collagen. The level of alkaline phosphatease had the same tendency. Conclusions FA could accelerate the differentiation of chondrocytes and mineralization of bone formation in certain concentration range in the process of endochondral ossification and thus influence may have relationship with KBD's pathological process.
