茄子花叶病是近年来山东分布较广的一种病害。发病植株表现斑驳、花叶、严重株叶片卷曲、矮化和畸形。从发病植株上分离到一病毒分离物(Smv—1)。可以通过汁液摩擦接种传播和由蚜虫非持久性传播。人工摩擦接种可侵染8科中的21种植物。系统侵染普通烟、心叶烟、黄瓜和番茄。局部侵染苋色藜、昆诺阿藜。此病毒的钝化温度为55~60℃10分钟,稀释限点为10^(-(?)~10^(-4)。体外存活期为5天(22C)。用差速离心法可以提纯该病毒,部分提纯的病毒最大紫外吸收峰为259nm,最小吸收峰为239nm,表现为典型的核蛋白吸收。A260/A280=1.68。SDS—不连续聚丙烯酰胺凝胶电泳解离病毒得到外壳蛋白亚基为一条多肽链,分子量约为25000道尔顿。琼脂免疫双扩散试验表明,Smv—1与黄瓜花叶病毒抗血清有阳性反应。用1.5%醋酸双氧铀负染。电镜观察部分提纯病毒粒子为球形(正二十面体)、直径为28~30nm。根据上述特征,Smv—1是黄瓜花叶病毒的一个株系。
The eggplant mosaic is a disease distributed widely in Shandong Province.A virus isolate (Smv—1) was obtained from severely infected plants which showed symptoms of mosaic, mottle, leafroll and distortion. The virus could be transmitted by sap inoculation to21 species of 8 families and by aphids (Myzus persicea and Aphis gossippi) in anon-persistant manner.Its thermal inactivation point was 55—60℃, the period of its life in vitro was 5 days(22℃), and the dilution end point was 10^(-3)—10^(-4).The virus could be partly purified by differental centrifugation. The virus preparation had a typical ultraviolet spectrum. The maximum UV absorption was 259nm and minimum 239nm. A260/ A280= 1.68. During SDS-discontinous polyacrylamide gel electrophoresis, Smv-1 revealed only one polypetide component of its coatprotein subunit, with a molecular weight about 25000 dolton.The virus gave a positive reaction to antiserum of CMV in the immune double diffusion test. Its particles were isometric, 28—30nm in diameter.The results show that Smv-1 is a strain of CMV.
