反相离子对色谱法测定人血浆中盐酸二甲双胍浓度 被引量:5

Determination of metformin in human plasma by reversed-phase ion-pair high performance liquid chromatography

摘要 目的建立高效液相色谱法分析血浆中盐酸二甲双胍浓度的方法.方法分析柱为Kromasil C18( 250mm×4.6mm,5μm),流动相为乙腈:磷酸缓冲液(内含少量十二烷基磺酸钠)40∶60,UV检测波长为233nm,采用高氯酸沉淀蛋白的方法,以吗啉双胍为内标.结果测定二甲双胍的线性范围为0.050~4.00μg·mL-1,日内精密度<8.18%,日间精密度<8.74%,方法回收率均>90%.结论本方法精密,准确,简便,适用于二甲双胍药动学的研究. OBJECTIVE A modified procedure has been established for the determination of metformin in human plasma.METHOD A Kramsil C_ 18 colum was used with a mobile phase of acetonitrile-phosphate buffer(60∶40),detected at 233nm.The samples were deproteinated with HClO_4,MLSG was used as the internal standard.RESULTS The calibration curve was linear over the range of 0.050~4.00μg·mL -1.The inter-day RSD was less than 8.18% and the intra-day RSD was less than 8.74%.The recovery was 92.05%~102.01%.CONCLUSION This method is simple with high sensitivity and good precision and is suitable for pharmacokinetic studies of metformin in humans.
