摘要
目的:研究关木通慢性肾损伤的功能改变和病理学变化与投药剂量、用药时间之间的关系。方法:取60只 SD 雌性大鼠随机分为4组,实验组 A、B、C 组按1g·kg^(-1)·d(-1)、2g·kg(-1)·d(-1)、4g·kg(-1)·d(-1)不同剂量分别给予关木通水煎液灌胃,正常对照组 D 组以等体积饮用水灌胃,分别于第6、12、14周末大鼠尾静脉取血,留取大鼠24小时尿液,测定相关肾功能指标,留取部分肾组织进行形态学检查,其余肾组织匀浆后测定相关指标。结果:在3个月观察期内《药典》法定剂量对大鼠肾功能及病理结构无不利影响,而剂量增大就会产生肾毒性,且与剂量、用药时间呈正相关。结论:关木通肾毒性是相对的。
Objective:To study on relation of functional and pathological changes of for chronic renal damage of Caulis Aristolochiane Manshuriensis with dose and lasting time of medication.Methods:60 SD female rats were randomly divided into 4 groups.Experimental group A,B,C were given intragastrically by lg·kg^(-1)·d^(-1),2g·kg^(-1)·d^(-1)and 4g·kg^(-1)·d^(-1)of Mutong Decoction,respectively,and the normal group(group D)by equal volume of potable water,and their blood samples were taken from the caudal vein at the ends of 6th,12th and 14th weeks,and 24h-urinay were taken for determination of indexes of renal functions,and a part of the renal tissue was taken for morphological detection and other renal tissue was homogenazed for determination of relative indexes.Results:The legal dose of pharmacopeia within 3 months did not have harmful effect on renal functions and pathological structures.but while the increase of the dose could induce renal toxity,whieh is related with the dose and lasting time of medication.Conclusion:The renal toxity of Caulis Aristolochiane Manshuriensis is relative.
