摘要
用HMBA作为诱导分化剂.研究了HMBA对人胃癌MGc80-3细胞的诱导分化效应,并采用免疫荧光技术观察了PKA-RⅡ与PKC-α亚型在细胞内分布的变化.细胞经5mmol/L HMBA处理后,细胞形态产生明显变化.细长突起明显增加,生长速度减慢,倍增时间延长.24小时,48小时,72小时的生长抑制率分别为20%.40.6%.54.8%.细胞在软琼脂上生长不成集落.细胞周期时相的分布发生变化、G_0+G_1期比对照组增加18.4%.而s期细胞比对照组减少14.8%.细胞内cAMP、DG水平也发生了显著变化.HMBA处理24小时后.细胞内cAMP水平上升62%.DG水平下降64.7.处理48小时后.cAMP水平上升76.2%.而DG水平下降的69.8%.以上结果充分表明了HMBA对人胃癌MGc80-3细胞的诱导分化作用.实验中还发现细胞经HMBA处理后.PKC-α分布于胞质,而PKA-RⅡ向胞核移位.提示HMBA对细胞的诱导分化作用与信号传递通路密切相关.
The effects of differentiation induced by HMBA in human gastric cancer cells were studies and the intracellular distribution of PKA - R Ⅱ and PKC - α were observed by immunocytochemical localization in the paper.After the cells were treated with 5mM HMBA, the morphological change and a decrease in cell generation were observed. Treating the cells for 24, 48 and 72h with HMBA, resulted in 20%, 40.6% and 54.8%, growth inhibition respectively. No colony was formed in soft agar. The cells were increased by 18.4% in G0+ G1 phase, and decreased by 14.8% in S phase, as compared with the control. HMBA caused an increased of cAMP level by 62%. decrease of DG level by 64.7 in MGc80-3 cells by 24h; Treated for 48h. cAMP level increased by 87% and DG level decreased by 69.8%. The results of this study demonstrate the effects of HMBA on growth inhibition and differentiation of human gastric cancer cell line MGc80-3.The PKC- a cytoplasm translocation and the PKA-R Ⅱ nuclear translocation after HMBA treatment suggested that the effects of the differentiation is closely related to signal transmission pathways.
